Interleukin-12 receptor subunit beta-2 (IL12RB2) is a type I transmembrane protein that serves as the essential signaling component of the high-affinity Interleukin-12 (IL-12) receptor complex [UniProt]. It heterodimerizes with the IL12RB1 subunit to bind IL-12, a cytokine critical for coordinating innate and adaptive immune responses [PubMed]. Expression of IL12RB2 is highly regulated and serves as a key marker for Th1 cell commitment, as its presence allows T cells to respond to IL-12 by producing interferon-gamma (IFN-gamma) via the JAK-STAT signaling pathway [NCBI]. In clinical contexts, mutations or deficiencies in IL12RB2 are associated with increased susceptibility to intracellular pathogens, such as mycobacteria, while its overactivity is implicated in chronic inflammatory and autoimmune diseases like Crohn's disease and multiple sclerosis [StatPearls]. Furthermore, IL12RB2 is frequently downregulated in various lymphoid and epithelial malignancies, where it is thought to function as a tumor suppressor by facilitating anti-tumor immunity [PubMed]. Therapeutic strategies often focus on modulating the IL-12 signaling axis, either by blocking the ligand or exploring direct receptor modulation, to treat chronic inflammatory disorders and certain malignancies [DrugBank].
IL12RB2 functions as the signaling subunit of the IL-12 receptor complex. Upon binding of the IL-12 cytokine (a heterodimer of p35 and p40) to the IL12RB1/IL12RB2 heterodimer, IL12RB2 mediates the recruitment and activation of Janus kinases, specifically JAK2 and TYK2. This activation leads to the phosphorylation of STAT4, which then translocates to the nucleus to promote the transcription of genes such as IFNG, thereby driving Th1-type immune responses and enhancing the lytic activity of NK cells [UniProt; PubMed].
