Overview

Interleukin-23 is a heterodimeric cytokine member of the IL-12 family, composed of IL-23p19 and IL-12p40 subunits[4][7][1][8]. It is secreted by activated dendritic cells, macrophages, monocytes, and other immune cells upon stimulation by antigens or pattern recognition receptors[4][8]. IL-23 exerts its biological effects via the IL-23 receptor, which activates the JAK2/TYK2/STAT3 signaling pathway, leading to Th17 cell maintenance and expansion, and release of proinflammatory cytokines crucial for defense against pathogens and the development of mucosal barrier immunity[2][4][8]. Aberrant activation or overproduction of IL-23 drives chronic inflammation and autoimmunity, especially in skin, gut, and joints[2][4][8]. Pharmacologic inhibitors of IL-23 or its receptor represent an important class of therapies for psoriasis, inflammatory bowel disease, and related disorders[2][5][6][8].
