Intestinal calcium and phosphate transporters are a group of membrane proteins and channels expressed by enterocytes responsible for the absorption of dietary calcium and phosphate across the intestinal epithelium. The main transporters for calcium include TRPV6 (an apical calcium channel), calbindin (an intracellular calcium-binding protein), and PMCA1b (a basolateral calcium ATPase). For phosphate, the principal transporter is the sodium-dependent phosphate transporter NaPi-IIb (SLC34A2). These systems are tightly regulated by hormones such as calcitriol (active vitamin D), parathyroid hormone, and fibroblast growth factor 23 to maintain systemic calcium and phosphate homeostasis critical for bone health and metabolic function. Disruption of transporter function leads to significant mineral balance disorders and is of therapeutic interest in metabolic bone and renal diseases.
