Overview

The ionizing radiation (IR) field within tumor tissue refers to the localized environment where high-energy particles or electromagnetic waves, such as X-rays or gamma rays, are applied to induce therapeutic damage to malignant cells (National Cancer Institute, 2023). This target is not a biological molecule but a physical stimulus that causes direct ionization of DNA and indirect damage through the radiolysis of water, generating reactive oxygen species (ROS) (StatPearls, Radiation Physics and Biology, 2023). These events lead to complex DNA lesions, most notably double-strand breaks (DSBs), which trigger cell death pathways such as apoptosis and senescence (PubMed, PMID: 28972013). Pharmacological agents often interact with this field as radiosensitizers, such as Cisplatin or 5-Fluorouracil, to enhance DNA damage, or as radioprotectors, such as Amifostine, to shield healthy tissues (American Cancer Society, 2022). The efficacy of the radiation field is significantly influenced by the tumor microenvironment, particularly oxygen levels, as molecular oxygen is required to fix or stabilize radiation-induced DNA radicals (PubMed, PMID: 11073131).
