Overview

JPX, also known as "JPX transcript, XIST activator (non-protein coding)", is a long non-coding RNA (lncRNA) located within the X-inactivation center (XIC) and functions as a key activator of XIST, the master regulator of X chromosome inactivation (XCI)[1][2][3]. JPX promotes XIST expression by interacting with architectural proteins such as CTCF, evicting repressors from the XIST promoter, and thus enabling chromosome-wide gene silencing in females[1][2]. JPX acts through both cis (local) and trans (distal) mechanisms to regulate XIST expression, and its transcript levels are critical for proper XCI[1][3]. In cancer contexts, JPX is frequently overexpressed and can act as an oncogenic factor by functioning as a competing endogenous RNA, sequestering specific microRNAs that regulate cell proliferation and invasion[2]. JPX does not code for a protein and is not a classical therapeutic target such as a receptor or enzyme, but plays a critical role in gene regulation and is implicated as a potential biomarker in oncology and developmental biology[2][3].
