Jun proto-oncogene, AP-1 transcription factor subunit (commonly known as c-Jun) is a protein encoded by the *JUN* gene in humans. It is a key component of the activator protein 1 (AP-1) transcription complex, typically forming heterodimers with proteins from the FOS family to regulate gene expression by binding specific DNA sequences. As one of the first discovered oncogenic transcription factors, c-Jun plays critical roles in controlling cell proliferation, apoptosis, differentiation, response to extracellular signals (such as growth factors and stress), and axon regeneration after nerve injury. The *JUN* gene is intronless and located at chromosome region 1p32-p31—a locus frequently altered in various malignancies including neuroblastoma. Overexpression or amplification of JUN has been observed in several aggressive cancers. While no direct small-molecule inhibitors are clinically approved for this target itself based on current data provided here, modulation often occurs through inhibition of upstream kinases like JNK that phosphorylate and activate c-Jun.
