The K99 fimbria, also known as F5 antigen, is a plasmid-encoded surface structure produced by certain enterotoxigenic Escherichia coli (ETEC) strains, enabling these bacteria to adhere tightly to the intestinal villi of neonatal calves, pigs, and lambs. This adhesion facilitates colonization of the distal small and large intestine, promoting secretory diarrhea through enterotoxin production and leading to severe, often lethal dehydration in affected animals aged 1-5 days.[1][2][3] K99 expression is influenced by growth conditions, with optimal production in minimal media at 37°C and reduced output in complex media or below 32°C, and it varies by the host strain's O antigen (e.g., higher in O101 strains).[1] The fimbriae are key virulence factors in the neonatal diarrhea complex, frequently co-occurring with pathogens like rotavirus or Cryptosporidium, and their presence correlates with profuse diarrhea and poor villous adherence in K99-negative strains.[2][3] Diagnostic tests like FASTest E.coli-K99 strips detect K99 in feces for rapid identification of carriers and targeted prophylaxis, highlighting its role as a primary etiological agent in veterinary infections worldwide.[3] While not a traditional molecular target like receptors or enzymes, K99 serves as a therapeutic target for vaccines and diagnostics aimed at preventing ETEC-mediated enteric diseases in livestock.[2][3]
