Overview

The KDM5D neighbour, Y-linked (KDNBY), commonly referred to by its clone name RP11-424G14.1, is a human-specific long noncoding RNA gene localized on the Y chromosome and predominantly expressed in male tissues. It has been implicated in regulating male-biased genetic factors and affects pathways linked to sexual dimorphism in immune response, cellular senescence, and metabolic regulation. Loss or knockdown of RP11-424G14.1 was shown to affect the NF-κB pathway and the expression of genes such as IGFBP3, suggesting roles in inflammation and aging. Unlike protein-coding therapeutic targets, KDNBY operates via modulation of gene expression and is not currently a direct target for pharmacological therapy, nor are there drugs known to interact with it. There is no evidence that KDNBY (RP11-424G14.1) is the cause of retinitis pigmentosa (RP11); this is a case of gene symbol overlap, not biological connection. While structurally and functionally distinct from classical receptors or enzymes, KDNBY has important roles in male-specific pathways and may contribute to sex-biased disease risk through regulation of gene networks, but lacks direct therapeutic utility at present.
