Keratin 32 (KRT32) is an acidic type I hair keratin, part of the intermediate filament protein family, predominantly expressed in the cuticular and basal layers of hair follicles and specific epithelial tissues such as skin and esophagus. KRT32 heterodimerizes with type II keratins to form the structural matrix of hair and nails. Beyond its structural role, KRT32 modulates keratinocyte behavior and skin immune responses, notably suppressing cell proliferation and inhibiting NF-κB signaling by promoting NEMO degradation. Damaging mutations in KRT32 lead to loss of anti-inflammatory function and excessive proliferation in keratinocytes, contributing to inflammatory skin disorders such as Pityriasis Rubra Pilaris. KRT32 is not currently targeted by approved drugs but emerging data support its potential relevance as a therapeutic or diagnostic target in inflammatory and proliferative skin conditions.
