Overview

Keratin, type II cytoskeletal 2 oral (KRT76) is a type II intermediate filament protein expressed largely in differentiated, suprabasal layers of stratified epithelia, including oral cavity, skin, and squamous stomach. KRT76 is essential for maintaining epithelial barrier integrity by regulating tight junction function, particularly through its interaction with Claudin 1; deficiency leads to skin barrier defects, impaired wound healing, hyperproliferation, and inflammation. Loss or downregulation of KRT76 is associated with certain inflammatory skin diseases such as psoriasis and correlates with poor prognosis in oral squamous cell carcinoma, suggesting a role in epithelial carcinogenesis and immune regulation. KRT76 is not currently considered a direct therapeutic target, and there are no known drugs that selectively interact with it. Its biological importance is highlighted in tissue homeostasis, cytokine regulation, and possibly in the modulation of antitumor immune responses.
