Overview

Keratin 36 (KRT36) is a type I hair keratin, an acidic, structural protein that heterodimerizes with type II keratins to form intermediate filaments in hair and nails[1][3][4]. It is encoded by the KRT36 gene, which is clustered in chromosome 17q12-q21[1][3][4]. KRT36 is expressed predominantly in the hair cortex, where it acts as a specific marker of terminal keratinocyte differentiation critical for the structural integrity and mechanical properties of hair[1]. It is also expressed in filiform papillae of the tongue, nail beds, and Hassal's corpuscles in the thymus, tissues that are subject to mechanical stress[2]. In sheep, KRT36 contributes to the physical properties of wool fibers. Downregulation of KRT36 has been observed in squamous cell carcinoma of the tongue, suggesting a possible role as a marker for pre-neoplastic change, although its exact role in tumorigenesis remains unclear[2]. Mutations or dysregulation in KRT36 have been associated with Familial Woolly Hair Syndrome[5]. KRT36 is not considered a classical receptor, enzyme, or transporter, nor is it a primary therapeutic target; rather, it is a structural component of epithelial appendages that provides mechanical resilience and differentiation-specific functions[1][2].
