Overview

Keratin 6B, type II cytoskeletal (KRT6B) is a structural intermediate filament protein abundantly expressed in differentiated epithelial tissues including the skin, hair follicles, nails, and specific glandular epithelia. KRT6B partners with type I keratins such as KRT16 and KRT17 to form dense intermediate filament networks, providing mechanical strength and resilience against physical stress. Mutations in KRT6B are linked to pachyonychia congenita type 2, presenting as nail dystrophy, painful palmoplantar keratoderma, and increased skin fragility. Altered expression plays roles in cancer cell migration and epithelial differentiation, but KRT6B does not act as a classic therapeutic target or signaling protein. Its biological importance is largely structural and diagnostic, rather than pharmacological.
