Overview

Killer cell immunoglobulin-like receptor, three Ig domains pseudogene 1 (KIR3DP1) is a member of the killer cell immunoglobulin-like receptor (KIR) gene family, located in the leukocyte receptor complex on human chromosome 19q13.4[5][6][8]. While most KIR family genes encode transmembrane glycoproteins involved in regulation of natural killer (NK) cell and T cell activity, KIR3DP1 is classified as a pseudogene because it is nonfunctional: it is not transcribed and lacks several functional domains typical for active KIRs, such as exons required for a full-length receptor protein[5][6][8]. KIR3DP1 is present in all haplotypes as a "framework" gene, structurally serving as a genomic reference point in the complex and polymorphic KIR region[6][8]. Unlike functional KIRs, KIR3DP1 does not bind ligands or transmit immune signals and consequently has no known direct biological or disease role, nor confirmed relevance as a biomarker, therapeutic target, or drug-interacting protein[5][6][8]. The KIR family as a whole plays a key regulatory role in immune responses by interacting with specific HLA class I molecules, but KIR3DP1's pseudogene status precludes a direct functional contribution[6][8].
