Overview

Lanosterol 14-alpha demethylase, also known as CYP51, is a cytochrome P450 enzyme that plays a critical role in the biosynthesis of ergosterol, an essential component of fungal cell membranes. This enzyme catalyzes the oxidative removal of the 14α-methyl group from lanosterol through a three-step monooxygenation process. The resulting product is further processed to form ergosterol, which regulates membrane permeability and rigidity in fungi. It is the primary target for azole antifungals, and resistance can arise through amino acid substitutions or increased efflux pump activity.
