Overview

Large ribosomal subunit protein mL51, mitochondrial (MRPL51), is a protein component of the 39S large subunit of the mitochondrial ribosome, encoded by the nuclear genome and imported into mitochondria, where it is essential for protein synthesis within the mitochondrion[3][4]. MRPL51 interacts with proteins such as OXA1L and functions in the biogenesis and insertion of membrane proteins in the mitochondrial inner membrane[1][2]. MRPL51 is crucial for mitochondrial DNA (mtDNA) integrity, respiratory growth, and redox homeostasis. Dysregulation of MRPL51, including its upregulation, has been implicated in the progression and aggressiveness of cancers such as lung adenocarcinoma, where it promotes cell proliferation, cell cycle progression, epithelial-mesenchymal transition (EMT), DNA repair, and invasion, partly through transcriptional activation by FOXM1[1][2]. High MRPL51 expression correlates with a poor prognosis in lung adenocarcinoma, suggesting its potential role as a biomarker. However, MRPL51 is not currently considered a therapeutic target, and no drugs are known to directly interact with it[1][2][3][4].
