Overview

Latent membrane protein 1 (LMP1) is the primary oncoprotein encoded by the Epstein-Barr virus (EBV) and is essential for the virus's ability to transform and immortalize B-lymphocytes (PubMed, UniProt) [1, 2]. It functions as a constitutively active mimic of the CD40 receptor, a member of the tumor necrosis factor receptor (TNFR) superfamily, and signals in a ligand-independent manner by recruiting cellular adapter proteins such as TNFR-associated factors (TRAFs) and TRADD (Wikipedia, PubMed) [2, 3]. This recruitment triggers a variety of downstream signaling cascades, including the NF-κB, PI3K/Akt, MAPK, and JAK/STAT pathways, which collectively promote cell survival, proliferation, and the inhibition of apoptosis (PubMed, Frontiers) [3, 4]. LMP1 is expressed in several EBV-associated malignancies, most notably nasopharyngeal carcinoma, Hodgkin's lymphoma, and post-transplant lymphoproliferative disorders, where it also contributes to immune evasion and changes in the tumor microenvironment (Frontiers, ecancer) [4, 5]. As a viral protein present on the surface of infected cells, LMP1 is a significant therapeutic target for the development of monoclonal antibodies, vaccines, and small molecule inhibitors (Frontiers, ecancer) [4, 5]. Current research explores targeting LMP1 directly or its downstream effectors, such as the Hippo pathway (YAP/TAZ), to treat EBV-driven cancers (PubMed, Frontiers) [1, 4].
