The Latent membrane protein 1 (LMP1) peptide–human leukocyte antigen (HLA) complex is a tumor-specific antigen target found on the surface of cells infected with Epstein-Barr virus (EBV), particularly in EBV-associated cancers such as nasopharyngeal carcinoma and Hodgkin lymphoma (Source: PubMed PMID: 33433515). LMP1 is a key viral oncogene that mimics CD40 signaling to drive oncogenesis, but its structure as a multi-pass transmembrane protein makes it difficult to target with standard monoclonal antibodies (Source: Blood, 2017). Instead, processed LMP1 peptides are presented by HLA molecules, creating a unique "TCR-like" target that can be recognized by engineered T-cell receptors or TCR-like antibodies (Source: Journal of Hematology & Oncology, 2022). This approach allows for highly selective targeting of malignant cells while avoiding healthy tissues that do not express the viral protein. Current therapeutic candidates, such as ET1402L1, utilize this mechanism to deliver potent cytotoxic effects specifically to EBV-positive tumor cells. However, the effectiveness of this strategy depends on the patient's HLA profile and the tumor's ability to maintain HLA expression, as downregulation is a common mechanism of immune evasion.
