Latherin-like protein (Fel d 8) (Fel d 8)

Target
Fel d 8
Molecular classification
Latherin-like protein, BPI/LBP/PLUNC superfamily, PLUNC family protein
01

Overview

Fel d 8 is a 24 kDa latherin-like protein that serves as an allergen in the domestic cat (Felis catus). It is primarily produced in the submandibular salivary glands and is found in cat saliva, which is subsequently transferred to the fur during grooming. As a member of the BPI/LBP/PLUNC superfamily, Fel d 8 possesses surfactant properties and shares significant sequence homology with horse allergens Equ c 4 and Equ c 5. Although it is considered a minor allergen compared to Fel d 1, it is recognized by specific IgE in approximately 19-20% of cat-allergic individuals and can trigger symptoms such as allergic rhinitis, conjunctivitis, and asthma. In the context of biotechnology, Fel d 8 is a key component in component-resolved diagnostics (CRD), which helps clinicians distinguish between primary cat sensitization and cross-reactivity with other mammalian species. Therapeutic strategies involving Fel d 8 focus on allergen-specific immunotherapy (AIT), where the protein is used to induce immunological tolerance in sensitized patients.

Mechanism of action

Induces IgE-mediated Type I hypersensitivity by cross-linking IgE-FcεRI complexes on mast cells and basophils, leading to the release of inflammatory mediators; used in allergen-specific immunotherapy (AIT) to induce immunological tolerance through T-cell and B-cell modulation.
