LIM homeobox protein 2 (LHX2) is a transcription factor belonging to the LIM homeobox family, characterized by two cysteine-rich LIM domains and a homeobox DNA-binding domain (UniProt P50458). It plays a critical role in the development of various organs, including the eye, forebrain, and hair follicles, by regulating the proliferation and differentiation of progenitor cells (NCBI Gene 9355). In the context of disease, LHX2 is frequently dysregulated; its overexpression is associated with several malignancies, such as pancreatic cancer and chronic myeloid leukemia, where it promotes tumor growth and metastasis (PubMed: 25605116). While LHX2 itself is a challenging target for small molecules due to its nature as a transcription factor, its mRNA is a potential target for nucleic acid-based therapies like siRNA or antisense oligonucleotides. Modulating LHX2 expression offers a therapeutic avenue for treating cancers and potentially certain developmental or regenerative conditions. Research has shown that silencing LHX2 mRNA can inhibit the growth of certain tumor types, making it a focus for experimental oncology (PubMed: 28415754).
