Overview

LNC-LBCS (LncRNA bladder and prostate cancer suppressor, hnRNPK interacting) is a long non-coding RNA that acts as a tumor suppressor in both bladder cancer and prostate cancer. In bladder cancer, particularly in bladder cancer stem cells, LNC-LBCS is downregulated and its reduced expression is associated with increased self-renewal capacity, chemoresistance, and poor prognosis. Mechanistically, LNC-LBCS binds directly to hnRNPK (heterogeneous nuclear ribonucleoprotein K) and EZH2 (Enhancer of Zeste Homolog 2), forming a regulatory complex that represses SOX2 transcription through histone methylation (H3K27me3 modification), thereby inhibiting stemness and drug resistance in bladder cancer cells. In prostate cancer, LNC-LBCS similarly functions by interacting with hnRNPK to suppress the translation of androgen receptor (AR) mRNA, limiting the emergence of castration resistance. Low expression of LNC-LBCS is correlated with worse clinical outcomes, and its restoration holds potential as a novel therapeutic strategy or biomarker in both bladder and prostate cancer. As a unique lncRNA, it does not encode a protein but mediates key regulatory axes at the post-transcriptional and epigenetic levels
