Long non-coding RNA, low expression in tumor (lncRNA-LET, also known as NPTN intronic transcript 1 or NPTN-IT1) is a long non-coding RNA gene with no protein-coding potential[3][4][5]. It is implicated in tumor biology, particularly in the regulation of hypoxia-induced pathways, epithelial-mesenchymal transition (EMT), cell migration, and apoptosis in various cancers, including lung squamous cell carcinoma and nasopharyngeal carcinoma[3][5]. lncRNA-LET interacts with the double-stranded RNA-binding protein NF90, promoting its degradation, which reduces the accumulation and stability of HIF-1α mRNA under hypoxic conditions. This in turn suppresses EMT-related proteins and inhibits cancer metastasis and invasiveness[1][5]. lncRNA-LET tends to be downregulated in tumors, and its overexpression is associated with inhibited EMT, reduced migration, and increased apoptosis of cancer cells[5]. It is studied primarily as a regulatory RNA involved in cancer progression, but is not itself a therapeutic target such as a receptor, enzyme, or transporter[3][4][5].
