Overview

Long non-coding RNA for association with Brahma (BRM) (lncBRM) is a regulatory long non-coding RNA that interacts with the chromatin remodeling ATPase SMARCA2 (also known as BRM), a core catalytic subunit of the SWI/SNF complex[1][3]. In liver cancer stem cells, lncBRM binds to SMARCA2 and inhibits the assembly of SMARCA2 (BAF)-containing SWI/SNF complexes, thereby promoting the formation of SMARCA4 (BRG1)-containing BAF complexes and activating YAP1-driven oncogenic signaling[1][3]. lncBRM is aberrantly overexpressed in liver cancer stem cells and correlates with more aggressive hepatocellular carcinoma phenotypes[3]. It plays a key role in determining cell fate by regulating the epigenetic landscape through chromatin remodeling, acting as a scaffold or decoy in macromolecular complexes[1][3]. There are no known small-molecule drugs that specifically target lncBRM.

Mechanism of action

Acts as a molecular scaffold/guide by associating with SMARCA2 to modulate chromatin remodeling complexes, switches the composition from SMARCA2-containing to SMARCA4-containing SWI/SNF complexes, thereby influencing YAP1 (oncogenic pathway) activity
