Overview

Lymphocyte antigen 6 complex, locus G6D (LY6G6D) is a cell surface protein belonging to the Ly-6/uPAR superfamily, characterized by a conserved three-fingered Ly6/uPAR (LU) domain with multiple disulfide bonds. LY6G6D is encoded within the MHC class III region on chromosome 6 and is transiently expressed during monocyte differentiation, but persistently expressed in mature granulocytes and circulating neutrophils. It plays a pivotal role in innate immunity, particularly in neutrophil function, including recruitment, migration, phagocytosis, ROS production, and bacterial clearance. LY6G6D expression is closely linked to immune function in neutrophils, and its downregulation is associated with immune senescence and reduced neutrophil activity in aging. In cancer, especially colorectal cancer, high LY6G6D expression is associated with increased risk of metastatic recurrence, and its expression is regulated by the JAK/STAT pathway, making it a potential therapeutic target. However, specific drugs targeting LY6G6D or a detailed molecular mechanism of drug action have not been described in the literature. LY6G6D is not known to interact with currently marketed drugs, and there are no notable safety concerns reported for this target at this time. Its expression level may serve as a biomarker for both immune senescence and cancer prognosis.
