Overview

Lymphocyte antigen 6 family member G6D (Ly6G) is a glycosylphosphatidylinositol (GPI)-anchored cell surface protein belonging to the Ly-6/uPAR superfamily [3, 11]. In murine models, Ly6G is a definitive marker for mature neutrophils and is essential for their recruitment and migration to inflammatory sites through interactions with β2-integrins [3, 7]. Although its expression on human neutrophils is negligible, Ly6G (LY6G6D) is significantly overexpressed in human colorectal cancer (CRC), specifically the microsatellite stable (MSS) subtype, where it contributes to an immunosuppressive tumor microenvironment [16, 20]. This selective expression makes it an attractive therapeutic target for antibody-based immunotherapies, such as bispecific T-cell engagers (TcEs) that redirect T-cell cytotoxicity against Ly6G-positive tumor cells [16, 22]. Additionally, anti-Ly6G antibodies are widely utilized in preclinical research to investigate the role of neutrophils in various pathologies, including infection and chronic inflammation [2, 13].
