Overview

Lymphocyte antigen 75 (CD205), commonly known as DEC-205, is a type I transmembrane C-type lectin receptor primarily expressed on dendritic cells (DCs), particularly the cDC1 subset, and thymic epithelial cells [1, 2]. It plays a critical role in the immune system by mediating the endocytosis of antigens and directing them to late endosomal/lysosomal compartments for efficient processing and presentation on both MHC class I and class II molecules [3, 8]. This unique trafficking pathway allows DEC-205 to facilitate cross-presentation, a process essential for initiating CD8+ T-cell responses against tumors and viruses [4, 9]. In the absence of inflammatory stimuli, DEC-205-mediated antigen uptake typically leads to immune tolerance, making it a potential target for treating autoimmune diseases [7, 12]. Conversely, when targeted alongside maturation signals like TLR agonists, it serves as a potent platform for cancer vaccines, such as CDX-1401 [14, 15]. Additionally, CD205 is overexpressed in various malignancies, including lymphomas and certain solid tumors, leading to the development of antibody-drug conjugates (ADCs) like MEN1309 that deliver cytotoxic agents directly to cancer cells [6, 11]. The receptor also functions as a recognition site for apoptotic and necrotic cells, contributing to the maintenance of peripheral tolerance [4, 13]. Recent structural studies have further elucidated its pH-dependent oligomerization and ligand-binding mechanisms, enhancing its utility as a therapeutic target [10, 18].
