Overview

Lymphotoxin-alpha1-beta2 (LTα1β2) is a membrane-bound heterotrimeric cytokine belonging to the tumor necrosis factor (TNF) superfamily, composed of one LTα and two LTβ subunits [PMID: 12891276]. It serves as the primary ligand for the lymphotoxin-beta receptor (LTβR), playing a fundamental role in the development and maintenance of secondary lymphoid organs and the organization of splenic architecture [UniProt: P01374, Q06643]. Beyond normal physiology, LTα1β2 is a key driver of chronic inflammation and the formation of tertiary lymphoid structures (TLS) in autoimmune conditions such as rheumatoid arthritis and Sjögren's syndrome [PMID: 16131644]. Therapeutic intervention typically involves the use of decoy receptors like baminercept (LTβR-Ig) or monoclonal antibodies to neutralize the heterotrimer, thereby inhibiting downstream pro-inflammatory signaling and lymphoid neogenesis [PMID: 21903606]. While targeting this pathway offers a novel approach to treating refractory autoimmune diseases, challenges include maintaining host defense mechanisms and managing the complex interplay within the TNF cytokine network.
