Lysophosphatidic acid receptor 6 (LPA6), also known as P2RY5, is a G protein-coupled receptor that serves as a high-affinity receptor for lysophosphatidic acid (LPA) [1, 2]. Unlike the classical LPA1-3 receptors, LPA6 belongs to the P2Y-like receptor family and is primarily expressed in the inner root sheath of hair follicles [1, 3]. It signals predominantly through G12/13 proteins to activate the Rho/ROCK pathway, which is essential for regulating the structural integrity and growth of the hair shaft [2, 4]. Mutations in the LPAR6 gene are strongly associated with autosomal recessive hypotrichosis and woolly hair syndrome, making it a key target for dermatological research [2, 5]. Additionally, aberrant expression of LPA6 has been observed in various malignancies, such as hepatocellular carcinoma, where it may contribute to tumor progression and cell motility [6]. While currently targeted primarily by endogenous ligands in research, LPA6 represents a promising therapeutic target for treating hair loss disorders and potentially certain cancers [1, 6].
