Overview

The 3' untranslated region (3'UTR) of the MAF BZIP transcription factor B (MAFB) mRNA is a pivotal regulatory domain that governs the post-transcriptional fate of the MAFB transcript (UniProt Q9Y5Q3). MAFB is a transcription factor essential for the terminal differentiation of macrophages and the functional maturation of pancreatic beta-cells (PubMed: 10551804). The 3'UTR serves as a scaffold for various microRNAs, such as miR-147 and miR-155, which bind to specific seed sequences to induce mRNA degradation or translational inhibition, thereby fine-tuning MAFB protein levels (PubMed: 21930763). In clinical contexts, dysregulation of MAFB is strongly associated with multiple myeloma, where chromosomal translocations often lead to its overexpression, and with metabolic diseases like diabetes (PubMed: 15150103). Therapeutic targeting of the MAFB mRNA 3'UTR is an emerging area of interest, utilizing antisense oligonucleotides (ASOs) or microRNA mimics to suppress MAFB expression in oncogenic or inflammatory states (PubMed: 25661213).
