Major histocompatibility complex, class II, DO beta chain (HLA-DOB) is a membrane-anchored beta chain that forms a heterodimer with an alpha (DOA) chain, collectively termed HLA-DO. This complex plays a regulatory role in the class II antigen presentation pathway of the immune system, being predominantly expressed in antigen-presenting cells such as B lymphocytes, dendritic cells, and macrophages. HLA-DO modulates the peptide loading activity of MHC class II molecules by inhibiting HLA-DM, thereby influencing the repertoire of antigens presented to CD4+ T cells and helping to shape immune responses. While critical to antigen processing and presentation, HLA-DOB is not a primary target for approved therapeutics, nor is it associated with known drug interactions or safety risks as a direct clinical target[1][2][3][5][6][7].
