HLA-A*02:01 is a specific allele of the HLA-A gene encoding a major histocompatibility complex (MHC) class I molecule, a heterodimer of a polymorphic heavy alpha chain (encoded by HLA-A) non-covalently associated with invariant beta-2-microglobulin. It presents short peptides (typically 8-10 amino acids) from intracellular proteins, such as viral or tumor antigens, on the cell surface for recognition by CD8+ T cells and natural killer cells via T cell receptors or KIRs. The peptide-binding groove features six pockets (A-F) with allotype-specific residues determining anchor preferences (e.g., L/M/I at P2 in pocket B, V/L/I/A at C-terminus in pocket F for HLA-A*02:01). It plays a pivotal role in adaptive immunity, with structural dynamics allowing peptide loading and presentation.
