Mammaglobin-A (MAM-A), also known as Secretoglobin family 2A member 2 (SCGB2A2), is a 10-kDa glycoprotein and a member of the secretoglobin superfamily (UniProt P15242). It is highly overexpressed in approximately 80% of primary and metastatic breast cancers, while its expression in normal tissues is restricted to the mammary, lacrimal, and salivary glands (PubMed: 15507677). The MAM-A2.1 peptide (sequence: LIYDSSLCDL) is a specific immunodominant epitope that is processed and presented by the HLA-A*02:01 major histocompatibility complex (MHC) class I molecule on the surface of breast cancer cells (PubMed: 22547526). This peptide-MHC complex is recognized by the T-cell receptors (TCRs) of CD8+ cytotoxic T lymphocytes, which then mediate the destruction of the tumor cells. Therapeutic strategies targeting this complex include DNA vaccines (e.g., pNMAM-A) and peptide-based vaccines designed to expand the population of MAM-A-specific T cells in patients (PubMed: 25437593). Clinical trials have demonstrated that targeting the MAM-A2.1 epitope is safe and can induce significant anti-tumor immune responses, making it a valuable target for precision immunotherapy in HLA-A2-positive breast cancer patients (ClinicalTrials.gov NCT00807781).
