Mammary tumor associated RNA 25 (MaTAR25) is a long non-coding RNA (lncRNA) that has been identified as a key driver of mammary tumor progression and metastasis (Chang et al., 2020, Nature Communications). Originally discovered in mouse models of breast cancer, it is highly upregulated in tumor tissues compared to normal mammary glands (Diermeier et al., 2016, Genes & Development). MaTAR25 functions by interacting with the purine-rich element-binding protein B (PurB) to regulate the transcription of genes like Tensin1 (Tns1), which is vital for cell-extracellular matrix interactions and migration (Chang et al., 2020). Its human ortholog, LINC01271, is also overexpressed in human breast cancer and correlates with poor patient prognosis (NCBI Gene ID: 100507410). Experimental therapeutic approaches using antisense oligonucleotides (ASOs) to knock down MaTAR25 have shown significant reduction in tumor growth and metastatic spread in vivo (Spector Lab, CSHL). Consequently, MaTAR25 represents a potential therapeutic target and a diagnostic biomarker for aggressive breast cancer subtypes.
