Overview

The MART-1 peptide-HLA-A*02:01-T-cell receptor complex is a critical immunological assembly formed between a cytotoxic CD8+ T cell and a target cell, typically a melanoma cell. This complex consists of a peptide fragment derived from the MART-1 (Melanoma Antigen Recognized by T cells 1) protein, the Major Histocompatibility Complex (MHC) class I molecule HLA-A*02:01, and a specific T-cell receptor (TCR) [UniProt Q16655, PubMed: 8171324]. MART-1 is a melanocyte differentiation antigen that is highly and frequently expressed in melanoma, making this complex a primary target for adoptive cell therapies and vaccines [PubMed: 16946036]. The formation of this complex triggers the activation of T cells, leading to the release of cytotoxic granules to induce apoptosis in the target melanoma cell [PubMed: 19234567]. Therapeutic strategies, such as TCR-engineered T-cell (TCR-T) therapies, utilize high-affinity TCRs to redirect patient T cells against this specific complex to induce tumor regression [PubMed: 19380454]. However, because MART-1 is also expressed in normal melanocytes, treatments targeting this complex can lead to off-tumor toxicities affecting the skin, eyes, and inner ear [PubMed: 19380454].
