MARVEL domain-containing protein 1 (MARVELD1) is a nuclear protein belonging to the MARVEL domain-containing family, which is typically associated with vesicle trafficking and membrane link functions (UniProt Q9BSK0). In the nucleus, MARVELD1 is involved in critical cellular processes including the regulation of the cell cycle, specifically the G1/S phase transition (PubMed: 23653360). It also plays a role in maintaining microtubule stability and has been shown to interact with pre-mRNA splicing factors, suggesting a role in RNA processing (PubMed: 21444676). MARVELD1 is widely recognized as a putative tumor suppressor, as its expression is frequently lost or significantly reduced in various cancers such as hepatocellular carcinoma, non-small cell lung cancer, and gliomas (PubMed: 19448941). This downregulation is often mediated by epigenetic mechanisms, particularly the hypermethylation of its promoter region (PubMed: 25670303). The loss of MARVELD1 is associated with increased tumor cell proliferation, migration, and chemoresistance, making it a potential prognostic biomarker. Currently, there are no FDA-approved drugs or clinical-stage candidates that specifically target MARVELD1 or its mRNA. However, research is ongoing into therapeutic strategies that could reactivate MARVELD1 expression, such as the use of DNA methyltransferase inhibitors. Its mRNA levels are also being explored as a diagnostic tool to monitor cancer progression and response to therapy. Overall, MARVELD1 represents a significant target for understanding tumor biology and developing future epigenetic-based interventions.
