Matrix metallopeptidase 2 (MMP2) mRNA is the transcript responsible for the synthesis of the MMP2 enzyme, a zinc-dependent endopeptidase also known as gelatinase A or 72 kDa type IV collagenase (NCBI Gene ID: 4313). This mRNA is a critical therapeutic target because the resulting protein plays a central role in the degradation of the extracellular matrix (ECM), particularly type IV collagen, which is a primary component of basement membranes (UniProt P08253). Overexpression of MMP2 mRNA is frequently observed in various malignancies and is strongly associated with increased tumor invasiveness, metastasis, and angiogenesis (PubMed: 25605234). By targeting the mRNA directly using technologies such as antisense oligonucleotides (ASOs) or small interfering RNAs (siRNAs), researchers aim to prevent the translation of the MMP2 protein, thereby inhibiting the pathological remodeling of the ECM (PubMed: 15150584). This approach offers a more specific alternative to broad-spectrum small molecule MMP inhibitors, which have historically faced challenges in clinical trials due to lack of selectivity and musculoskeletal toxicity (PubMed: 12195160).
