Overview

Matrix metallopeptidase 7 mRNA (MMP7 mRNA) is the messenger RNA transcript that encodes the MMP7 enzyme, also known as matrilysin (NCBI Gene: 4316). MMP7 is a member of the matrix metalloproteinase family and is unique because it lacks a hemopexin-like domain, making it one of the smallest members of the family (UniProt: P09237). The mRNA is frequently overexpressed in various epithelial cancers, including colorectal and pancreatic carcinomas, where it facilitates tumor invasion and metastasis by degrading the extracellular matrix and activating other proteases (PMID: 15623931). In addition to oncology, MMP7 mRNA levels are significantly elevated in the lungs of patients with idiopathic pulmonary fibrosis (IPF), serving as a critical biomarker for disease severity and progression (PMID: 22430196). Therapeutic strategies targeting MMP7 mRNA, such as siRNA or antisense oligonucleotides, aim to silence the production of the protein at the pre-translational level. This approach allows for the inhibition of MMP7's pathological roles in tissue remodeling and cell signaling before the enzyme is even synthesized. Research has demonstrated that silencing MMP7 mRNA can reduce the invasive potential of cancer cells and modulate fibrotic pathways in preclinical models. As a target, MMP7 mRNA offers a high degree of specificity compared to small molecule inhibitors that often struggle with selectivity among the MMP family.
