Overview

Matrix metalloproteinase-13 (MMP13) mRNA is the transcript encoding the MMP13 enzyme, a member of the matrix metalloproteinase family involved in the breakdown of extracellular matrix components. The protein product, also known as collagenase 3, is uniquely characterized by its high specificity for degrading type II collagen, the primary structural protein in articular cartilage (UniProt P45452). Under physiological conditions, MMP13 expression is minimal, but it becomes highly upregulated in diseases such as osteoarthritis and rheumatoid arthritis, where it drives joint destruction (PubMed PMID: 23810431). Beyond joint diseases, MMP13 mRNA expression is associated with tumor progression and metastasis in various cancers due to its role in tissue remodeling. Targeting the mRNA transcript itself, rather than the resulting protein, is an emerging therapeutic strategy utilizing RNA interference (RNAi) or antisense oligonucleotides (ASOs). This approach aims to achieve high specificity and long-lasting suppression of the enzyme, potentially overcoming the musculoskeletal toxicity observed with non-selective small molecule MMP inhibitors (PubMed PMID: 31463456). Experimental therapies often utilize specialized delivery systems, such as lipid nanoparticles or polymer-based carriers, to transport the RNA-targeting agents into chondrocytes within the avascular cartilage environment.
