Overview

The MXRA5 peptide–HLA-A*02:01 complex is a specific peptide-major histocompatibility complex (pMHC) class I target utilized in cancer immunotherapy. Matrix-remodeling-associated protein 5 (MXRA5), also known as adicuin, is a secreted and cell-surface protein involved in extracellular matrix assembly and remodeling that is frequently overexpressed in various solid tumors, including non-small cell lung cancer, glioblastoma, and colorectal cancer, while maintaining restricted expression in normal adult tissues (PMID: 22496204, 21135219). In this complex, a specific immunogenic peptide derived from the MXRA5 protein (such as the MXRA5-001 peptide) is processed and presented on the cell surface by the HLA-A*02:01 allele. This presentation allows the tumor cells to be recognized by the immune system, specifically by cytotoxic T lymphocytes (CTLs) carrying cognate T-cell receptors (TCRs). Therapeutic strategies targeting this complex include multi-peptide cancer vaccines like IMA910 and IMA950, as well as the development of TCR-based adoptive cell therapies and bispecific molecules designed to redirect T-cell toxicity toward MXRA5-expressing malignant cells (Immatics, 2023).
