Overview

This entry corresponds to a pseudogene of the MDM2 family, not the canonical E3 ubiquitin-protein ligase Mdm2 which plays a pivotal role in regulating p53, cell cycle, and tumorigenesis[1][2][3][4]. Pseudogenes are segments of DNA that resemble functional genes but typically do not produce functional proteins due to mutations or lack of regulatory sequences. The canonical Mdm2 protein is a validated therapeutic target in oncology due to its interaction with and regulation of p53, but the pseudogene does not contribute to these biological functions and is not considered a therapeutic target in itself. The functional MDM2 gene/protein (not the pseudogene) is a crucial oncogene, an E3 ubiquitin ligase that inhibits and promotes proteasomal degradation of p53, thus regulating cell cycle, apoptosis, and genomic stability. It is upregulated in various cancers and is a bona fide therapeutic target, with small molecule inhibitors like nutlins in clinical development[1][2][3][4]. However, these properties do not apply to the pseudogene encoded by ENSG00000223691.
