Overview

The gp100:154-162 peptide is a specific immunogenic epitope derived from the melanocyte protein PMEL, commonly known as gp100 (UniProt: P40967). This nine-amino acid sequence (KTWGQYWQV) is a naturally processed peptide that is presented on the surface of melanoma cells by the Human Leukocyte Antigen (HLA) class I molecule, specifically the HLA-A*02:01 allele (PubMed: 11158039). As a tumor-associated antigen, it serves as a critical target for cancer immunotherapy, particularly in the development of peptide-based vaccines and T-cell receptor (TCR) engineered therapies (NCI Drug Dictionary). By administering this peptide, clinicians aim to stimulate a robust cytotoxic T-lymphocyte (CTL) response that specifically recognizes and destroys gp100-expressing melanoma cells (PubMed: 10427150). While gp100 is also expressed in normal melanocytes, its high expression in melanoma makes it a viable target, though it can lead to autoimmune side effects like vitiligo (PubMed: 15150571). Clinical studies have frequently evaluated this peptide in combination with cytokines like interleukin-2 or immune checkpoint inhibitors to enhance anti-tumor efficacy (PubMed: 21631324).
