Overview

The MAGE-A1 peptide presented on HLA-A*01:01 complex refers to the complex of a short peptide fragment derived from the melanoma antigen gene-A1 (MAGE-A1) bound within the peptide-binding groove of the human leukocyte antigen class I molecule HLA-A*01:01. This peptide-MHC (pMHC) complex is expressed on the surface of certain cancer cells and is recognized by T cell receptors (TCRs) on cytotoxic T lymphocytes, triggering immune responses against tumor cells expressing the MAGE-A1 antigen. The structure of this complex has been resolved, revealing the precise interactions between the peptide, HLA-A*01:01, and both TCRs and specialized antibodies with TCR-like specificity. The MAGE-A1-HLA-A*01:01 complex is a validated therapeutic target in TCR-engineered T cell therapies and TCR-like antibody approaches for cancer immunotherapy, particularly for tumors expressing cancer-testis antigens such as MAGE-A1. Biomarker strategies and therapy selection depend on both tumor MAGE-A1 expression and patient HLA typing. Safety considerations include the risk of targeting non-malignant tissues that may express the antigen and the necessity for precise antigen and HLA restriction to avoid unwanted immune reactions.
