Melanoma-associated antigen 1 peptide–HLA-C*07:02 complex (MAGE-A1–HLA-C*07:02)

Target
MAGE-A1–HLA-C*07:02
Molecular classification
Peptide–major histocompatibility complex (pMHC), Antigen, Class I MHC complex, Tumor antigen complex
01

Overview

The **MAGE-A1 peptide–HLA-C*07:02 complex** is a tumor-associated antigenic structure composed of a peptide from the MAGE-A1 protein bound by the HLA-C*07:02 class I molecule and presented on the cell surface. This complex is specifically recognized by cytotoxic T lymphocytes through their T cell receptors or by engineered therapeutics such as TCR–T cells and TCR-like antibodies. MAGE-A1 peptides presented by HLA-C*07:02 constitute targets for cancer immunotherapy, especially in patients whose tumors and immune systems express this combination. The identification, structural validation, and therapeutic targeting of this complex offer promise for precision oncology, although clinical application is constrained by HLA genotype, peptide sequence specificity, and potential immunological risks.

Mechanism of action

Targeted immune cell killing: TCRs or CARs engineered to recognize the MAGE-A1 peptide–HLA-C*07:02 complex bind and initiate cytotoxicity of cancer cells expressing this target. Antibody-mediated recognition: TCR-like antibodies can bind the complex and recruit immune effectors.

Safety considerations
