Overview

The MAGE-A1 peptide–MHC complex is a highly specific tumor target consisting of a peptide fragment derived from the Melanoma-associated antigen 1 (MAGE-A1) protein presented on the cell surface by Major Histocompatibility Complex (MHC) Class I molecules, most commonly HLA-A*01. MAGE-A1 is a member of the cancer-testis antigen (CTA) family, which is characterized by high expression in various malignancies—including melanoma, lung, and liver cancers—while remaining silent in normal adult tissues, except for the immune-privileged germ cells of the testis. Because the testis lacks MHC Class I expression, the MAGE-A1–MHC complex is effectively tumor-specific, making it an ideal candidate for T-cell receptor (TCR) based therapies. Therapeutic strategies targeting this complex include TCR-engineered T-cells (TCR-T) and bispecific T-cell engagers designed to redirect the immune system to recognize and eliminate MAGE-A1-positive cancer cells. Clinical development focuses on ensuring high affinity for the specific peptide-HLA combination while avoiding cross-reactivity with similar peptides found in vital organs.
