Overview

Melanoma-associated antigen 10 (MAGE-A10) is a member of the MAGE-A subfamily of cancer-testis antigens (CTAs). Under normal physiological conditions, its expression is restricted to immune-privileged sites such as the testes and placenta, but it is aberrantly overexpressed in various malignancies, including melanoma, lung, and bladder cancers. This restricted expression pattern makes MAGE-A10-derived epitopes, specifically short peptides presented by Human Leukocyte Antigen (HLA) molecules, highly attractive targets for cancer immunotherapy. Therapeutic strategies targeting MAGE-A10 primarily involve adoptive cell transfer using T-cell receptor (TCR) engineered T-cells. These engineered cells are designed to recognize specific MAGE-A10 peptide-HLA complexes on the surface of tumor cells, triggering a potent cytotoxic immune response. Clinical development has focused on HLA-A*02 restricted epitopes, such as the GLYDGMEHL peptide. While promising, the development of these therapies requires rigorous screening to avoid cross-reactivity with similar sequences in healthy tissues, which could lead to severe adverse events.
