Melanoma-associated antigen 10 (MAGE-A10) presented by HLA-A*02:01 is a peptide-major histocompatibility complex (pMHC) that serves as a specific target for cancer immunotherapy, particularly T-cell receptor (TCR) engineered T-cell therapies [1.2.1, 1.3.2]. MAGE-A10 is a member of the cancer-testis antigen (CTA) family, which is typically expressed only in the immune-privileged sites of the testes and placenta but is aberrantly overexpressed in various malignancies, including melanoma, non-small cell lung cancer, and urothelial carcinoma [1.1.1, 1.1.4]. The specific 9-mer peptide GLYDGMEHL derived from MAGE-A10 is processed intracellularly and presented on the cell surface by the HLA-A*02:01 allele [1.2.1, 1.5.2]. This presentation allows the immune system, or engineered therapeutic T-cells, to recognize and eliminate cancer cells while sparing most healthy tissues [1.3.3]. Therapeutic agents like Adaptimmune's ADP-A2M10 (SPEAR T-cells) utilize affinity-enhanced TCRs to bind this complex with high specificity [1.2.1, 1.2.4]. However, development challenges include the risk of off-target cross-reactivity with similar peptides in vital organs and the requirement for patients to possess both the HLA-A*02:01 genotype and MAGE-A10-positive tumors [1.5.1, 1.5.3].
