Overview

Membrane protein, palmitoylated 3 (MAGUK p55 subfamily member 3), abbreviated as MPP3, is a membrane-associated scaffold protein that belongs to the MAGUK (membrane-associated guanylate kinase homologs) family[1][3]. MAGUKs are key organizers of multiprotein complexes at intercellular junctions and regulate cellular architecture by interacting with the cytoskeleton. MPP3 contains a conserved SH3 domain and a catalytically inactive guanylate kinase (GK) domain, which play a crucial role in structural assembly and protein-protein interactions, especially in adherens junctions and signaling complexes[1][4]. MPP3 also interacts with the PI3K signaling pathway and contributes to cell spreading and stabilization of certain cell surface receptors[1]. While associated with intellectual disability and microdeletion syndromes, MPP3 is not currently a direct target for pharmacological intervention.
