Overview

Membrane protein, palmitoylated 5 (MAGUK p55 subfamily member 5) (PALS1), encoded by the MPP5 gene, is a scaffold protein of the MAGUK family critical to the assembly and maintenance of cell junctions in epithelial tissues. It contains PDZ, SH3, GUK, and L27 domains, allowing interaction with tight junction proteins such as PATJ and Crumbs homologues, thereby forming complexes that orchestrate cell polarity and tight junction integrity. PALS1 regulates apical-basal polarization, junctional protein trafficking, and neural/retinal development. Dysfunction or reduced expression is linked to developmental abnormalities, such as retinal degeneration, and general disruption of epithelial architecture. It does not have established direct roles in drug targeting or serve as a primary biomarker, but participates in pathways relevant for tissue organization and cancer suppression via MAGUK family function.
