Memory T-lymphocytes are a specialized subset of antigen-experienced T cells that persist long-term after an initial immune challenge (StatPearls, 2023). They are characterized by their ability to mount a faster and more potent immune response upon re-exposure to a specific pathogen or antigen compared to naive T cells (Nature Reviews Immunology, 2013). These cells are broadly classified into central memory (Tcm), effector memory (Tem), and tissue-resident memory (Trm) subsets based on their expression of markers like CCR7 and CD62L and their localization (PubMed, PMC4406145). While essential for long-term immunity against infections and cancer, dysregulated memory T cells can drive chronic inflammatory conditions and autoimmune diseases (NIH, 2018). Pharmacological intervention typically targets specific receptors on these cells, such as CD52 or CD25, to either deplete them or modulate their activation and proliferation (Journal of Clinical Investigation, 2015).
Therapeutic agents modulate memory T-lymphocytes by targeting surface proteins (e.g., CD52, CD25) to induce cell depletion, or by interfering with intracellular signaling pathways (e.g., calcineurin inhibition, mTOR inhibition) to suppress activation and proliferation (Journal of Clinical Investigation, 2015). Additionally, some drugs sequester these cells in lymphoid tissues by targeting trafficking receptors like S1P (Nature Reviews Drug Discovery, 2013).
