Merozoite surface protein 1 (MSP1) is a high-molecular-weight glycoprotein and the most abundant surface antigen found on the merozoite stage of the malaria parasite Plasmodium falciparum (UniProt: P04932). It is synthesized as a 190-200 kDa precursor during the late stages of parasite development and undergoes two essential rounds of proteolytic processing (PubMed: 10858498). The final cleavage event occurs just prior to or during erythrocyte invasion, leaving a 19 kDa C-terminal fragment (MSP1-19) anchored to the parasite membrane as it enters the host cell (PubMed: 15596776). MSP1 is critical for the parasite's life cycle because it mediates the initial attachment and subsequent entry into human red blood cells, which is the stage responsible for the clinical symptoms of malaria. Because of its essential role and high visibility to the host immune system, MSP1 has been a primary focus for the development of blood-stage malaria vaccines (NIH: PMC3347517). Therapeutic strategies primarily involve inducing antibodies that bind to MSP1 to sterically hinder invasion or facilitate parasite clearance through opsonization. However, the high degree of genetic polymorphism in the MSP1 gene across different parasite populations remains a significant challenge, often leading to strain-specific immunity that limits the broad efficacy of vaccine candidates (PubMed: 17037517).
