Metadherin (MTDH), also known as Astrocyte elevated gene-1 (AEG-1), is a scaffold protein and potent oncogene that is overexpressed in over 20 different types of human cancer. It plays a pivotal role in promoting tumor progression, epithelial-mesenchymal transition (EMT), metastasis, and resistance to chemotherapy by modulating multiple signaling pathways, including NF-κB, PI3K/Akt, and Wnt/β-catenin. The 3' untranslated region (3'-UTR) of the MTDH mRNA is a critical regulatory domain that governs the stability and translation of the transcript through interactions with microRNAs and RNA-binding proteins. Recent therapeutic advances have focused on targeting this 3'-UTR with small molecules, such as the compound C-10, which binds to specific structural motifs within the region to promote the degradation of the MTDH mRNA. By reducing MTDH protein levels, these agents aim to inhibit the interaction between MTDH and its partners like SND1, thereby suppressing the pro-metastatic and chemoresistant phenotypes of aggressive cancers.
